分泌抗—HBe单克隆抗体杂交瘤细胞株的建立

Establishment of hybridoma cell lines secreting monoclonal antibodies against hepatitis virus e antigen

用ＨＢｅＡｇ阳性血清纯化ＨＢｅＡｇ，免疫ＢＡＬＢ／Ｃ小鼠脾细胞与小鼠骨髓瘤细胞进行融合，反复克隆化培养，最终获得４株分泌抗－ＨＢｅ单克隆抗体的杂交瘤细胞株。对其中两株进行了鉴定。其细胞培养上清中的ＥＬ／ＩＳＡ效价分别为１：８００和１：５００，诱生同属小鼠腹水中的ＥＬＩＳＡ效价均为１：１０００，特异性试验表明，两株杂交瘤分泌的抗体为特异性抗体，小鼠Ｉｇ亚类鉴定，两株抗体均为ＩｇＧ１亚类球蛋白。细胞染色体数为９５～１１０条。结果证明，此细胞株为特异性分泌抗—Ｈｋｅ单克隆抗体细胞株。

Four hybridoma cell lines secreting mouse monoclonal antibodies (McAbs) against hepatitis virus e antigen (HBeAg) were established with the fussion of mouse myeloma cells and the spleen cells of the BALB/C mouse immunized with purified HBeAg.Two of the four cell lines were identified.The titers of McAbs in the culture supernatants were 1:800 and 1:500,and in ascites from mice bearing intraperitoneally transplanted hybridomas were 1:1000.McAbs were specific for HBeAg. The subclasses of both McAbs were IgG1.The chromosome numbers of the hybridoma cell were 95～110.It suggested that the cell lines might be specific ones which secreted the monoclorlal anti-HBe antibodies.
