分泌抗—HBe单克隆抗体杂交瘤细胞株的建立

摘要 用HBeAg阳性血清纯化HBeAg,免疫BALB/C小鼠脾细胞与小鼠骨髓瘤细胞进行融合,反复克隆化培养,最终获得4株分泌抗-HBe单克隆抗体的杂交瘤细胞株。对其中两株进行了鉴定。其细胞培养上清中的EL/ISA效价分别为1:800和1:500,诱生同属小鼠腹水中的ELISA效价均为1:1000,特异性试验表明,两株杂交瘤分泌的抗体为特异性抗体,小鼠Ig亚类鉴定,两株抗体均为IgG1亚类球蛋白。细胞染色体数为95~110条。结果证明,此细胞株为特异性分泌抗—Hke单克隆抗体细胞株。 Four hybridoma cell lines secreting mouse monoclonal antibodies (McAbs) against hepatitis virus e antigen (HBeAg) were established with the fussion of mouse myeloma cells and the spleen cells of the BALB/C mouse immunized with purified HBeAg.Two of the four cell lines were identified.The titers of McAbs in the culture supernatants were 1:800 and 1:500,and in ascites from mice bearing intraperitoneally transplanted hybridomas were 1:1000.McAbs were specific for HBeAg. The subclasses of both McAbs were IgG1.The chromosome numbers of the hybridoma cell were 95~110.It suggested that the cell lines might be specific ones which secreted the monoclorlal anti-HBe antibodies.
