摘要
目的观察脑外伤后不同数量移植神经干细胞的疗效是否存在不同,并找出受伤后进行干细胞移植的适宜量。方法成年大鼠制成脑外伤模型在伤后24h进行神经干细胞移植,移植量分别为20万,100万,200万。伤后1w动物运动神经功能评分后,处死取脑,行病理及免疫组化染色。结果伤后1w,接受100万单位干细胞移植的治疗组与损伤组以及其他治疗组(20万,200万)相比呈现明显的运动功能改善。结论移植量并不是越大越好,中等量移植效果比较理想,是适宜的移植量。
Objective To find out if transplanting neural stem cells with different quantity after traumatic brain injury(TBI) would make different outcome, hence , to decide the appropriate quantity for transplantation. Methods Transplanted neural stem cells (NSCs) with different quantity of 20 thousand, 1 million, and 2 million ceils at 24 hours after traumatic brain injury of rats. The motor function of rats was assessed. The techniques of immunohistochemistry were applied to define the NSCs, differentiation and neuroprotection, Results Brain injured animals that have received NSCs transplants with the quantity of 1 million had marked and significant improved motor function scores compared with other groups. Conclusion Too much quantity of neurotransplantation was not always effective, The quantity of middle level would be appropriate,
