纳米磷酸三钙复合人工骨的程序外DNA合成试验 被引量:1

摘要 目的:从DNA的角度来探讨纳米磷酸三钙/胶原复合人工骨的遗传毒理学特性。借以了解此复合人工骨的生物相容性,为其在骨缺损修复领域的临床应用提供依据。方法:将纳米级磷酸三钙(-βTCP)同胶原复合,制成纳米-βTCP/胶原复合人工骨,并制备浸提液,采用程序外DNA合成检测法(UDS试验),以生理盐水为阴性对照,测试大鼠原代肝细胞的每分钟放射计数(CPM)值。结果:材料浸提液的CPM均值为1262,与阴性对照组(814)比较无显著性差异(P>0.05),与阳性对照组(6734)比较差异显著(P<0.01)。结论:纳米磷酸三钙/胶原复合人工骨不会引起大鼠肝细胞程序外DNA合成增加,提示此人工骨不会引起DNA损伤。 Objective: To study the inherited toxicology of nano-tricalcium phosphate/collagen (N-TCP/Co) artificial bone from the aspects of DNA as to study the bio-compatibility and safety of it for the clinical use of bone' s reconstruction. Method: Compound the N-TCP and Collagen to produce the N-TCP/Co artificial bone and it's diffusion. Test the diffusion through kIDS test. The nature saline is the control, The marker of observation is the radio counting per minute (CPM) in rat primary hepatocyte. Result:The CPM of the experimental group is 1262 which has not significant difference from that of the negative control group (814) and has significant difference from that of the positive control group (6734). Conclusion: The nano-tricalcium phosphate/collagen (N-TCP/Co) artificial bone will not cause the increase of the unscheduled DNA synthesis of rat primary hepatocyte which shows that this artificial bone will not cause DNA damage.
