Effects of Benzalkonium Chloride on Human Tenon's Fibroblasts in Tissue Culture

摘要 目的观察苯扎氯铵对体外培养的人Tenon囊成纤维细胞的增殖的影响。方法体外培养人眼Tenon囊成纤维细胞。配制苯扎氯铵溶液,使其浓度依次为20、2~(2×10-7)μg/mL,采用噻唑蓝(MTT)比色法检测不同浓度的苯扎氯铵作用下细胞的增殖,并在光学显微镜下观察细胞的形态变化。结果较高浓度组(浓度≥0.2μg/mL)的苯扎氯铵对人Tenon囊成纤维细胞存在明显的杀伤作用,20μg/mL时细胞几乎全部死亡,A值与阴性对照相比明显减少(P<0.01),浓度为2和0.2μg/mL时细胞形态变化明显。较低浓度组(浓度<0.2μg/mL),A值和细胞形态较阴性对照均没有明显变化(P>0.05)。比较不同时间下的剂量效应曲线,4和24h没有显著差异(P>0.05)。结论苯扎氯铵0.2μg/mL及以上浓度对体外培养的成纤维细胞具有细胞毒性,低浓度时则对细胞增殖没有明显的影响。 Purpose To observe the effect of benzalkonium chloride on the proliferation of cultured human Tenon's capsule fibroblasts (HTCFs) in vitro. Methods Human Tenon's capsule fibroblasts were cultured in vitro. Solution of benzalkonium chloride was prepared and serially diluted to the concentration ranging from 20μg/mL, 2 μg/mL to 2×10^-7 μg/mL. Fibroblasts growth was measured with MTT assay method. Morphological changes were observed under phase contrast microscope. Results Benzalkonium chloride in higher concentration groups (at concentration of 0.2μg/mL or higher) take had effect on cultured human Tenon's capsule fibroblasts. When the concentration of benzalkonium chloride was 20μg/mL, the cells were almost dead and the A values were significantly decreased (P〈0.01). 2μg/mL and 0.2μg/ mL benzalkonium chloride induced obvious morphological changes. There were no significant changes of A values (P〉0.05) and morphological changes in lower concentration groups (lower than 0.2μg/mL). Comparing the dose-response curves of benzalkonium chloride after different time, there was no statistical difference in proliferation of HTCFs between 4 hours and 24 hours(P〉0.05). Conclusions Preservative benzalko-nium chloride has toxic effect at concentration of 0.2μg/mL or higher, while there is no influence on the proliferation of HTCFs at the lower concentrations.
