二硫代氨基甲酸吡咯烷预防腹腔粘连及其机制探讨

摘要 目的:探讨腹腔内注射核因子κB(NF-κB)抑制剂二硫代氨基甲酸吡咯烷(PDTC)对腹腔粘连的预防作用及其作用机制。方法:健康成年SD雄性大鼠96只,随机分成对照组、盲肠型肠粘连模型组(模型组)及PDTC组(术前1 h腹腔注射PDTC)(n=32)。各组大鼠随机分成4个亚组,每组8只,分别于术后第1、3、5、7天开腹观察腹腔粘连情况,进行粘连程度分级评定;同期抽取大鼠下腔静脉血,测定血清中肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)的含量。结果:腹腔粘连程度分级评定,PDTC组和模型组分别为(1.94±1.16)、(2.63±1.16),明显高于对照组(0.22±0.42,P<0.01);与模型组相比,PDTC组腹腔粘连程度显著减轻(P<0.05)。与模型组相比,PDTC组血清IL-6、TNF-α含量显著降低(P<0.05,P<0.01);对照组与PDTC组无显著差异。结论:腹腔粘连早期,细胞因子TNF-αI、L-6明显升高;PDTC可通过降低血清TNF-αI、L-6的含量来预防腹腔粘连。
