酶法测定1-磷酸葡萄糖的含量

摘要 建立反应液中1-磷酸葡萄糖(G-1-P)的测定方法.分别在样品中加入PGluM(EC5.4.2.2)、G6P-DH(ECl.1.1.49)和氧化型辅酶Ⅱ(β-NADP),通过测定还原型辅酶Ⅱ(β-NADPH)吸光值的变化(△A)来确定样品中G-1-P的含量.G-1-P在0~400 mg/L范围内与△A呈线性关系,r2=0.999 8,其平均回收率为100.88%(RSD=0.52%).该法快速、灵敏、准确. A rapid and specific enzymic-sepectrophotometric method was described for determination of D - glucose - 1 - phosphate ( G - 1 - P) in fermentation liquid by using phosphoglucomutase ( PGIuM EC5.4.2.2 ) ,glucose -6 - phosphate dehydrogenase (G6P-DH EC1.1.1.49) and NADP, respectively. The mass concentration of G - 1- P is linear with the change of absorption of β-NADPH (AA) in the range of 0 - 400 mg/L ( r2 = 0. 999 8 ) , and the recovery of this method is 100.88% (RSD =0.52% ). The method is snecific,rapid and accurate.
