摘要
卡介克蟹注射液是卡介苗(BCG)的提取物,我们应用MTT法已证实其对K562细胞的能量代谢有直接的抑制作用。本组实验用极限稀释法研究了卡介克蟹对K562肿瘤干细胞单个克隆细胞的影响;并观察卡介克蟹作用K562后,其细胞超微结构的改变。结果显示:浓度为1/8~1/128的卡介克蟹对K562肿瘤干细胞有直接的抑制作用,其克隆存活分数为0.46~0.82;高浓度的卡介克蟹(浓度为1/8)作用K56210h后,电镜可见肿瘤细胞质中出现大量空泡结构。提示:高浓度卡介克蟹对肿瘤细胞有直接的杀伤作用。
It is known that BCG has an effect against tumor, but the side effects are serious. In the present experiment, the anti-tumor effect 0f Ka Jie Ke xie (KJKX), an extract of BCG, was studied on K_562 tumor stem cells in vitro by limiting dilution assay and electron microscopy (EM). The results showed that after 14 days culture of K_562 cells with 1/8~1/128 KJKX, the colongenic survival fractions were 0.46~0.82. EM demonstrated a large number of vacuolar structures in the cytoplasm of K_562 cells after incubation with KJKX of highet conceutration (1/8) for 10h, These findings suggest that higher-concentration KJKX is directly cytotoxic to tumor cells.
