鲁梅克斯k-1杂交酸模及几种野生酸模的遗传多态性分析

摘要 利用RAPD技术对鲁梅克斯k - 1杂交酸模的 3个连续繁育世代和几种野生酸模进行遗传多态性分析。鲁梅克斯k - 1杂交酸模的 3个连续世代之间不存在多态性位点差异 ,3个世代是稳定遗传的 ;不同种和采自不同地点的同一个野生种之间存在着多态性位点差异。实验表现在合格种的 12个单株间具有明显的遗传多态性 ,采用混合取样可消除品种内的个体遗传差异性 ,较好地代表群体遗传特征。从OPA组选出 7个能扩增出清晰且稳定带谱的引物 ,7个引物共检测出 4 5个位点 ,DNA片段的大小在 30 0~ 2 5 0 0bp之间。 The genetic polymorphisms in the three continued generations of Rumex k - 1 ( Rumex patientia × R. tianschanicus, cv. Rumex k- 1) and native Rumex spp. were analysised by using random amplified polymorphic DNA markers (RAPD). Seven primers selected from twenty primers amplified 45 RAPD fragments. The length of most amplified fragments ranged from 300 bp to 2 500 bp, There were not polymorphic loci among the three continued generations of Rumex k- 1. The genetic stability was good in the three continued generations of Rumex k- 1. There were abundant genetic polymorphisms among different native species in Rumex and different individuals within the same variety from different local except for the genetic differentiation among 12 plants of the certified seed of Rumex k- 1.
