芦荟粗多糖对体外培养人表皮细胞增殖作用的研究 被引量:2

摘要 目的 研究芦荟粗多糖对体外培养人表皮细胞增殖活性、细胞内Ca^2+水平及细胞LDH漏出率的影响。方法 应用不同浓度(75, 150, 300, 600, 1 200 mg ·L^-1)芦荟粗多糖作用于表皮细胞,观察计算生长曲线和细胞增殖指数,并测定细胞内Ca^2+水平和LDH漏出率。结果 芦荟粗多糖组表皮细胞培养2 d后增殖速度明显增快,d 3~5细胞增殖指数呈剂量依赖性上升。与对照组比较,600和1 200 mg·L^-1芦荟粗多糖组表皮细胞内Ca^2+水平升高(P〈0.05),而这两组LDH漏出率则下降(P〈0.05)。结论 芦荟粗多糖具有促进体外表皮细胞增殖和保护表皮细胞的作用,并可能通过调节细胞内Ca^2+来发挥其促进细胞增殖作用。 Aim To investigate the effects of aloe coarse polysaeeharid on proliferation and the intraeellular eal cium level and the leakage rate of lactate ase (LDH) of the epidermal cells in vitro. dehydrogenMethods The epidermal cells were treated with aloe coarse polysaccharid of 75, 150, 300, 600 and 1 200 mg·L^-1 respectively. The proliferation of epidermal cells was observed interms of the proliferation curve and the population doubling/day (PD/d). The concentration of intracellular calcium and the leakage rate of LDH were also measured. Results In aloe coarse polysaccharid group, a dose-dependent and statistically significant (P 〈0. 05 -0. 01 ) promotion of epidermal cells proliferation was observed after 3, 4 and 5 days of incubation. Compared with the control group, the levels of intracellular calcium were significantly increased and the leakage rate of LDH was markedly decreased in the treatment group treated with 600 and 1 200 mg · L^- of aloe coarse polysaccharid ( P 〈 0. 05 ). Conclusion Aloe coarse polysaccharid may have protective effect and regulate intracellular calcium which results in the proliferation of the epidermal cells.
