桂枝芍药知母汤对Ⅱ型胶原诱导性关节炎大鼠血清肿瘤坏死因子-α、白细胞介素1β活性的影响

Effect of Guizhishaoyaozhimu Decoction on Serum Activity of TNF-α,IL-1β in Rats with CⅡ-Induced Arthritis

目的观察桂枝芍药知母汤对类风湿性关节炎(RA)的治疗作用,并探讨其作用机理。方法以Ⅱ型胶原(CⅡ)诱导的免疫性关节炎(CIA)大鼠为RA动物模型,通过测量其关节肿胀程度、关节炎指数观察桂枝芍药知母汤大、中剂量组的治疗效果;采用ELISA法测定大鼠血清中肿瘤坏死因子-α(TNF-α)活性,小鼠胸腺细胞法检测血清中白细胞介素1β(IL-1β)活性,探讨其作用机理。结果桂枝芍药知母汤大、中剂量组均可使大鼠踝关节肿胀程度和关节炎指数明显减少,与模型组比较差异显著(P<0.01);同时发现模型组大鼠血清TNF-α含量及IL-1β活性明显升高,治疗组起下调作用,接近正常组水平,与模型组比较差异显著(P<0.01)。结论TNF-α、IL-1β异常增高,与RA发病密切相关。桂枝芍药知母汤可以降低CIA大鼠血清中异常增高的TNF-α、IL-1β浓度,从而抑制或控制RA病情发展。

Objective To investigate the therapeutic effect and mechanism of rheumatoid arthritis, Methods Arthritis of rat induced by type Ⅱ collage was used as rheumatoid arthritis model, Selling of ankles of rats was measured and arthritis index was assessed to observe the therapeutic effect of Guizhishaoyaozhimu （GZSY） Decoction. The activity of TNF-α in serum was investigated with ELISA method. And the activity of IL-1 β in serum was investigated by thymocyte cell proliferation. Results Compared with that of model group, the swelling of GZSY group and arthritis index were relieved significantly. The activity of TNF-α and IL-1 β in model group were significant higher than those in normal group. But in GZSY group, the activity of those cytokines was decreased significantly. Conclusion GZSY has an effect on immune arthritis induced by type Ⅱ collage, and the decrease of TNF-α and IL- 1 β in serum may be one of its mechanisms.
