摘要
目的探讨实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis,EAE)非敏感品系Wistar大鼠加用不同剂量牛型结核杆菌卡介苗(mycobacterium bovis bacillus calmette-guerin,M.bovis BCG)对EAE模型诱导的作用。方法将Wistar大鼠分为EAE1、EAE2、EAE3、EAE4共4组,然后分别用含4、8、10、12 mg M.bovis BCG/mL完全福(氏)佐剂(CFA)加豚鼠脑脊髓匀浆(GPSCH)制备不同浓度的完全抗原,分别免疫诱导Wistar大鼠EAE。观察不同剂量M.bovis BCG对诱导EAE模型发病情况、各项临床指标的影响,同时取脑和脊髓制成石蜡切片,行HE染色观察组织形态变化,行单核细胞趋化蛋白-1(MCP-1)mRNA原位杂交(ISH)计数其阳性细胞百分率。结果EAE3、EAE4组的发病率、体重减轻、临床症状评分、MCP-1 mRNA阳性细胞数显著高于EAE1、EAE2组(均P<0.05)。结论M.bovis BCG浓度的高低影响Wistar大鼠EAE的发病率。
Objective To induce experimental autoimmunc encephalomyelitis (EAE) model in un-sensitive Wis:ar rats with different doses of mycobactcrium bovis bacillus calmctte-guerin (M. bovis BCG). Methods Wistar rats are divided randomly into groups EAE1, EAE2, EAE3, EAE4 for animal model. The 4 mg, 8 mg, 10 mg, 12 mg M. boris BCG/mL complete frcund's adjuvant (CFA) and guinea pig spinal cord homogenate were made the different concentrations of complete antigen, the different groups were immunized by subcutaneous injection in the foodpads with 0. 4 mL different concentrations of complete antigen and 0. 05 mL pertusis toxin. The brain and spinal cords were havested, embedded in paraffin, performed on 5-μm-thick. The expression of MCP-1 were determined by the in situ hybridization (ISH) method according to the instruction. Results The incidence of groups EAE weight change, clinical scorc and thc exprcssion of monocyte chcmokine protein-1 (MCP-1) mR- NA in EAE3 and EAE4 are statistically significantly highcr than thosc in EAE1, EAE2 (P〈0.01). Conclusions The concentrations of M. bovis BCG may influcncc incidence of EAE.
