摘要
目的:探讨异位子宫内膜细胞中整合素α5的表达及性激素对其的影响。方法:对18例卵巢子宫内膜异位症(EM)患者的异位子宫内膜组织行体外细胞培养并传代。传代培养中细胞分为非药物干预组(对照组)、雌二醇(E2)组、安宫黄体酮(P)组、米非司酮(RU486)组、三苯氧胺(TAM)组5组,细胞收获后用逆转录-聚合酶链反应技术检测各细胞组中整合素α5mRNA的表达。结果:整合素α5在E2组及P组的表达水平分别为1.415±0.286及1.304±0.170,与对照组的1.082±0.069相比,差异具有统计学意义(P<0.05),而TAM组及RU486组中整合素α5表达水平(分别为0.882±0.013及1.304±0.170)明显低于对照组(P<0.05)。结论:整合素α5表达受雌、孕激素调控,E2、P可能通过上调整合素α5的表达而促进EM粘附的发生,TAM、RU486可能通过拮抗E2、P或抑制异位内膜细胞分泌整合素5α分子而延缓EM的发展,从而起到治疗作用。
Objective To study the expression of integrin α5 in cultured human endometriotic cells and its regulation by steroidal honnones. Methods Eighteen samples of ectopie endometrium were cultured in vitro. The cultured endometfiotie cells were divided into five groups: control group, E2 group, P group, TAM group and RU486 group. The expression of integrin α5 mRNA of each group were determined by reverse transcript polymerase chain reaction method. Results The expression of integrin α5 mRNA in E2 and P group was 1.415 ± 0.286 and 1.304 ± 0. 170 respectively,which were significantly higher than that of control group( 1.082 ± 0. 069, P 〈 0.05). In TAM and RU486 group, it was 0. 882 ± 0.013 and 0. 866 ± 0.054 respectively , which was significantly lower than that of control group ( P 〈 0.05). Conclusions Estradiol and progesterone may regulate the expression of integrin α5 , Estradiol and progesterone increase the integrin α5 mRNA in ectopie endometrium, and promote the adhesion leading to endometriosis. Tamoxifen and mifepriston decelerate occurrence of endometriosis by way of resisting estradiol and progesterone or inhibiting eetopie endometrial cells from secreting integrin α5.
