当归补血汤对内皮细胞和骨髓造血细胞的增殖和分化作用

EFFECTS OF DBD TO ENDOTHELIAL CELLS AND HEMOPOIETIC CELLS IN MICE MARROW ON THE PROLIFERATION AND DIFFERENTIATION

目的:探讨当归补血汤对内皮细胞及骨髓造血细胞的刺激增殖和诱导分化作用。方法:在培养的内皮细胞及骨髓造血细胞半固体和液体培养体系中加入不同浓度的当归补血汤,采用流式细胞术、双抗夹心法检测其对内皮细胞及骨髓造血细胞增殖的影响。结果:当归补血汤能够促进内皮细胞由G1期进入S期及细胞因子的分泌,但对BFU-E,CFU-E,CFU-GM和CFU-M K集落产率的提高及细胞表面表达CD 13和CD 71的细胞数量没有统计学上的意义。结论:在体外,当归补血汤能够促进内皮细胞的增殖及细胞因子的分泌,但未发现促进骨髓造血干细胞增殖与分化的作用。

Objective： To investigate the action of danggui buxue decoction （DBD） on the proliferation and differentiation of endothelial cells and bone marrow hemopoietic stem cells. Methods： By culturing the endothelial cells and adding DBD with different concentration to the medium, ELISA analysis, Western blotting, Flow cytometry analysis and hemopoietic colony assay were used. Results： DBD can promote the eneothelial cells from G1 phase to S phase and the production of cytokine in endothelial cells （IL-6 and GM-CSF, etc. ）, but can not increase the proliferation and differentiation of hemopoietic cells in mice marrow. Conclusion： In vitro, the effects of DBD on the proliferation and differentiation of hemopoietic cells depend on regulation of secreting cytokines in endothelial cells by DBD.
