针刺预处理对脑缺血再灌注大鼠中枢下丘脑室旁核ACTH样蛋白表达的影响 被引量:7

EFFECT OF ACUPUNCTURE PRECONDITIONING ON ACTH-PROTEIN EXPRESSION OF HYPOTHALAMUS PARAVENTRICULAR NUCLEUS IN RATS WITH CEREBRAL ISCHEMIC REPERFUSION INJURY

摘要 目的:采用免疫细胞化学技术,探讨肾俞、百会穴针刺预处理对脑缺血再灌注模型大鼠不同时期中枢下丘脑室旁核ACTH样蛋白表达的影响。方法:以双侧肾俞、百会穴电刺激为针刺预处理方式,以颈动脉引流法行全脑缺血再灌注造模。术后分6h、24h、72h 3个时间段取脑,脑组织恒冷箱连续冠状切片,免疫细胞化学ABC反应,图像分析系统行下丘脑室旁核ACTH阳性免疫面积(stain area)、平均吸光度和(den-sity)检测。结果:各组stain area和density值呈同步变化。从术后6h开始,其它各组ACTH样蛋白表达较正常对照组显著增多,术后24h达高峰(P<0.05),随后逐渐回落。在3个不同时间段,各组ACTH样蛋白的表达存在显著差异(P<0.05)。结论:肾俞、百会穴针刺预处理对正常及脑缺血再灌注模型大鼠下丘脑室旁核ACTH样蛋白表达具有明显影响;可能是肾俞、百会穴针刺预处理发挥抗脑缺血再灌注损伤作用的机理之一。 Objective:To explore the effect of acupuncture preconditioning(AP) at Shensuh(BL23) and Baihui(DU20) on ACTH-protein expression of hypothalamus paraventricular nucleus in rats with cerebral ischemic reperfusion injury at different time points.Methods:AP at Shenshu(BL23) on both sides and Baihui(DU20) of the rats were electroacupunctured.Rat model of cerebral ischemic reperfusion was made with the method of carotid artery drainage.The brain was taken out respectively 6h,24h and 72h after the operation.The brain tissue was sectioned in box with constantly cold temperature.After ABC reaction of immunocytochemistry,the slices were analyzed under image analysis system to get the value of ACTH-positive stain area and density.Results:ACTH-positive substance was deeply brown.There were deeply stained cell nuclei in different areas of the brain with obvious nucleolus.There were positive nerve fibers like a string of beads distributing in thalamencephalon and hypothalamus.The results of image analysis system showed that values of stain area and density in different groups changed in-phase 6h after the operation,expression of ACTH in sham operation group,AP control group,model control group of brain ischemic reperfusion,AP and brain ischemic reperfusion group was significantly higher than that in normal control group,the expression reached acme 24h after the operation(P<0.05),later it declined.At three different time points,there was significant difference among the values of stain area and density of the groups.Conclusion:AP at BL23 and DU20 had obvious effect on ACTH protein expression of hypothalamus paraventricular nucleus in normal and brain ischemic reperfusion rats;AP at BL 23 and DU20 regulates ACTH expression in hypothalamus paraventricular nucleus so as to affect substance composition and metabolism of the nerves and maintain the stabilization of the ischemic reperfusion nerves,central nervous system and the whole body,which may be one of important approaches to resist brain ischemic reperfusion injury.
