2,3,7,8-TCDD对培养乳鼠心肌细胞内游离钙离子浓度影响的研究 被引量:3

摘要 目的测定培养SD大鼠乳鼠心肌细胞内游离钙离子浓度[Ca2+]i,观察2,3,7,8四氯二苯并二噁英(2,3,7,8tetrachlorodibenzopdioxin,以下略为TCDD)对心肌细胞[Ca2+]i的影响。方法将实验动物分为0.05、0.5、5、10μg/kg4个染毒组,采用Fluo3/AM同时加入PluronicF127作为细胞内游离钙离子的荧光探针,负载培养的心肌细胞,应用共聚焦激光扫描显微镜(CLSM)技术检测空白组及染毒组乳鼠心肌细胞[Ca2+]i。结果静息时,心肌细胞[Ca2+]i为(77.68±12.00)nmol/L。染毒组心肌细胞的[Ca2+]i显示不同程度的升高,且呈剂量依赖性。结论应用Fluo3/AM和PluronicF127结合CLSM技术可以精确的反映细胞内游离钙离子浓度,TCDD可能通过某种机制增加心肌细胞[Ca2+]i,从而对乳鼠心脏功能产生一定损伤。 Abstract: Objective To measure free Ca^2+ concentration of cultured cardiac cells of neonatal rats, and observe the effects of 2, 3, 7, 8-TCDD on [ Ca^2+]i. Methods The pregnant SD rats were randomly assigned to receive 2, 3, 7, 8-TCDD injection at the dose of 0, 0, 05, 0.5, 5, 10 μg/kg respectively on gestation day 8. The cardiac myocytes of 5-day neonatal rat were cultured, and Fluo-3/AM and Pluronic F-127 were loaded as Ca^2+ sensitive probe. The changes of intracellular Ca^2+ was observed by confocal laser scanning microscopy (CLSM). Results Undr the quiescent condition, the value of Ca^2+ of cardiac muscle was (77.68± 12.00) nmol/L in 2, 3, 7, 8-TCDD group at the dose of 0 μg/kg (control group), while the [ Ca^2+] i was (77.16 ± 13.20), (93.62 ±22.95), (110.99 ±25.96), (135.96 ±21.08) nmol/L in the 2, 3, 7, 8-TCDD treated group at the dose of 0.05, 0.5, 5, 10 μg/kg respectively. There was no statistical difference in [Ca^2+]i between control group and 0.05 μg/kg TCDD group, but significant difference between control group and other TCDD treated groups in which [Ca^2+]i increased in a concentration-dependent manner. Gonclusion CLSM system can accurately reflect cytosolic concentration of free Ca^2+ labeled by Fluo-3/AM and Pluronic F-127. TCDD may increase [Ca^2+]i, then make some impairment to the heart function of the neonate rats.
