摘要
目的 :研究阳离子脂质体介导nm2 3-h1质粒体内转染腺样囊性癌 (ACC)的可行性。方法 :将腺样囊性癌细胞株体外培养后移植入 2 4只裸鼠体内。ACC瘤体长至直径 1cm左右时 ,用脂质体包裹nm2 3-h1质粒进行瘤体内注射 ,每次注入质粒及脂质体复合物 0 1ml (其中含质粒约 2 0 μg/ml) ,其中 6只裸鼠共注射 2次 ,6只裸鼠注射一次 ,6只作空白对照。 6只裸鼠一次注射 0 2ml质粒及脂质体复合物。注射后 2、 3、 7天处死动物 ,获取肿瘤标本作常规组织学及免疫组化研究。结果 :瘤体内注射 3天后肿瘤细胞内即有nm2 3-h1表达 ,7天后表达增加 ,肿瘤细胞外间质增加 ,2次注射较一次注射、多针道注射较单一针道注射有更多的细胞呈阳性表达 ,但阳性表达范围小 ,效率低 ,不具备临床实施的可能性。
Objective: To investigate the feasibility of pla+smid nm23 - h1 transfeetion on high metastatic potential adenoid cystic carcinoma (ACC- M) cell line mediated by cationic lipid. Methods: ACC- M cells ( 10^6) were implanted in the maxillofacial region ill each of 24 BALB/c nude mice. After the tumor growth to lcm in diatneter, 0.1nd Lipofctin- nm23 - h1 plasnfid Complex (20μg/ml) were injected intratumorally in 6 mice, 6 mites injected for twice, 3 days after the first injection; 6 mice as plamid- blank control, and another 6 mice were injected 0.2ml complex. 2, 3, 7 days after the injection. The mice were killed and the specimens for HE stain and inununohistological chemistry study. Results; nm23 - h1 expression initiated in the tumor cells 3 days +after the complex injection, 7 days later, the expression level increases accompanying with extraeellular matrix increase, twice injection and multiple channel injection will gain better nm23 - h1 expression than only one injection and single- chalmel injection respectively. But the results gave little prospect for further clinical trial in such a maturer.
