摘要
目的分离培养人子宫平滑肌细胞,为研究人子宫平滑肌细胞生理和病理等方面提供了一个十分有用的实验模型。方法采用胶原酶消化法培养人子宫平滑肌细胞,可获得95%以上具有正常代谢活性的平滑肌细胞,对存活的子宫平滑肌细胞经倒置显微镜观察和免疫组化染色对其进行鉴别。结果平滑肌细胞接种4h后开始贴壁,6~7d汇合成片,所培养的细胞在倒置显微镜下呈梭形和典型“峰-谷”样生长,α-Actin免疫组化染色强阳性。结论采用本法体外培养的人子宫平滑肌细胞生长良好,纯度高,可用于人子宫平滑肌细胞生理和病理等方面的研究。
Objective Human myometrial smooth cells(hMSMCs) were isolated and primarily cultured, which provided a useful experimental model for study the physiology and pathology of hMSMCs. Methods Collagenase was used to iso- late hMSMOs. More than 95 % of the dissociated cells exhibited initial viability ( typan blue dye exclusion and the ability of adhesion). The surviving cells were observed by phase contrast microscope and verified by imrnunohistoohemistry staining. Results Some of hMSMCs became adherent after 4h cultivation, and reached confluence with 6- 7 days. The cells grew with spindle-shape and showed typical “hill - valley”pattern under phase contrast microscope. Histoohemical staining for smooth muscle α- Actin showed positive reaction in more than 97% attached cells. Conclusions The hMSMCs cultured in this experiment survive well, with highly purity, and can serve the further studies of physiology and pathology.
