摘要
目的细胞粘附现象与移植免疫排斥的关系愈来愈为人们所关注,本研究通过对细胞间粘附分子-1(ICAM-1)在大鼠同种肝移植物中的表达情况的分析,探讨ICAM-1与急性肝移植排斥反应的关系。方法采用免疫组织化学技术ABC法检测大鼠移植肝组织ICAM-1的表达。结果30例移植肝组织中,均见肝细胞膜、胆管上皮及炎性细胞有大量的ICAM-1表达,中央静脉内皮细胞呈低表达,而正常大鼠肝细胞无一例有ICAM-1表达。结论大鼠肝移植后ICAM-1在移植肝组织中的诱导表达可能是产生急性排斥的主要步骤,其机制可能是通过促进移植物浸润细胞的粘附而加剧移植排斥反应的进程。
Objective To investigate expression of Intercellular adhesion molecule-1(ICAM-1) on liver tissues from rats with acute rejection and compared with those of normal liver. Methods Lavis and ACI Rats were used as donor and recipient. Liver transplantation was performed under the ether. Immunohistochemistry was performed for analysis of liver samples from Rats with acute rejection and normal Rats. The relationship between ICAM-1 expression and rejection was analyzed. Results The results showed that there was stong expression of ICAM-1 on bile duct, sinusoid endothelium, perivenular hepatocytes and graft infiltrating cells in Rats with acute rejection, but ICAM-1 expression on normal liver is negative. Conclusions The induction of ICAM-1 on liver tissues may be an important step in the development of the inflammatory response of rejection. The upregulated ICAM-1 expression in the liver allograft may accelerate graft rejection by augmenting ad- hesiveness of graft infiltrating cells. ICAM-1 expression on liver tissues is of indicative of refection.
