摘要
目的:探讨血红素氧合酶-1在延迟性异种心脏移植排斥反应中的表达及意义.方法:建立NIH Wistar颈部异位心脏移植模型,分别于移植前、移植后1h、6h、12h、24h、48h和72h切取移植心脏,应用RT-PCR、Western Blot检测HO-1mRNA、HO-1蛋白表达并检测HO 1酶活性;同时比较钴原卟啉(CoPP)诱导HO-1对延迟性异种心脏移植排斥反应的影响.结果:移植心脏均有HO-1表达,HO-1mRNA(t=2.5170,P<0.05)、HO-1蛋白表达(t=2.3702,P<0.05)及酶活性(t=2.246,P<0.05)移植术后24~48h表达到达高峰;CoPP诱导的HO-1明显延长了移植心脏的存活时间(t=4.74442,P<0.001).结论:HO-1表达在延迟性异种心脏移植后24~48h到达高峰;CoPP诱导的HO-1能延长延迟性异种心脏移植的存活时间.
Objective :To investigate the expression and significance of heine oxygenasc- 1(HO- 1 ) in delayed cardiac xenograft rejection in NIH - Wistar. Methods:The model of cervical heterotopic heart transplantation in NIH - Wistar was established. The hearts were harvested 1,6, 12,24,48 as well as 72 hours before and after transplantion respectively. HO- ImLRNA and HO- 1 protein were examined by RT - PCR and Western Blot, HO- 1 enzymatic activity was examined. The survival time of transplanted heart, in which HO - 1 was induced by cobalt protoporphyrin(CoPP), was compared with the control. Results : HO - 1 was expressed in transplanted heart. The expression of HO- 1 mRNA( t = 2. 5170, P 〈 0.05 ), protein( t = 2. 3702, P 〈 0.05 ) and enzymatic activity( t = 2. 246, P 〈 0.05 ) in heart 24 and 48 hours after transplantion were higher than those in other groups,Conclusion : The expresskin peak of HO- 1 in NIH- Wistar cardiac transplantation is 24-48 hours after transplantion. HO- 1 induced by CoPP can prolong the survival time of delayed xenograft cardiac transplantation.
