摘要
目的:观察抵抗素(Resistin)对肝细胞糖代谢关键酶葡萄糖激酶(GK)和磷酸烯醇式丙酮酸激酶(PEPCK)活性的影响及剂量效应。方法:正常大鼠的肝细胞,以不加Resistin与加不同剂量的Resistin培养24h,用6-磷酸葡萄糖脱氢酶偶联比色法、乳酸脱氢酶偶联比色法检测不同剂量Resistin对GK和PKPCK的影响。结果:Resistin浓度为10nM/L、50nM/L时,GK活性与空白对照组无明显差异,PEPCK活性明显升高(P<0.05);当Resistin浓度上升为100nmol/L后,GK活性较空白对照组明显下降(P<0.01),而PEPCK活性升高更明显(P<0.01)。结论:抵抗素浓度升高使大鼠肝细胞GK活性下降,PEPCK活性升高,从而抑制糖酵解促进糖异生,升高血糖。上述两种酶的活性与抵抗素浓度呈现不同的剂量依赖关系。
Objective. To observe the effect of resistin on activity of gluccoskinase (GK) and phosphoenolpyruvate carboxykinase (PEP(K) in cultured rat liver cells. Methods.Rat liver cells were incubated with resistin of different doses (ranging from 10nM/L to 100nM/L). Controled liver cells were invubated without resistin. Glucose- 6- phosphate dehydrogenase couple chromatometry and lactate dehydrogenase couple chromatometry were used to determine activity of GK and PEP(K. Results ; Compared with the control group, treated with resistin of 10nM/L and 50nM/L for 24h had no significant effect on activity of GK, but activity of PKP(K increased significantly ( P 〈 0.05). At the dose of 100nM/L, resistin inhabited activity of GK markedly ( P 〈 0.01 ), while activity of PKPCK increased more significantly ( P〈0.01 ). Conclusion .The increase of Resistin leads to the decrease of activity of GK and increase of activity of PEPCK in rat liver cells, hence inhabite glycolysis and promote glyconeogensis and increase glucose as well. The activity of two enzymes shows dose dependent relation with resistin.
