组织芯片检测肝癌组织中GATA4及甲胎蛋白的表达及二者相关性 被引量:5

摘要 目的:探讨转录因子GATA4和甲胎蛋白(AFP)在肝癌中的表达、意义及二者相关性。方法:应用免疫组化ABC法检测29个肝癌和37个癌旁组织组织芯片中GATA4和AFP的表达。结果:肝癌组织中GATA4和AFP的阳性率分别为96.5%和82.7%,GATA4主要在核表达,胞质也有表达;癌旁组织中GATA4和AFP的阳性率为分别为94.5%和78.3%,GATA4主要在胞质表达,胞核也有表达。而癌和癌旁组织中AFP均在胞质表达。癌和癌旁组织中GATA4和AFP表达既有相关性又有统计学差异。GATA4的表达,癌组织较癌旁组织高,低分化癌较高分化癌高。结论:GATA4和AFP均在肝癌和癌旁组织表达,二者相关性较好,但GATA4比较敏感。GATA4的转录活性与肝癌细胞的分化程度密切相关。 Objective: To study the expression and relationship of GATA4 and AFP in hepatocarcinoma. Methods; The expression of GATA4 and AFP was detected by immunohistochemistry with hepatocarcinoma tissue microarray (29 carcinoma and 37 adjacent tissue). Results: The positive rates of GATA 4 and AFP in carcinoma were 96. 5% and 82. 8%, respectively, with more GATA 4 expression in nuclei than in plasma; the positive rates of GATA 4 and AFP in adjacent tissue were 94. 5 % and 78. 3%, respectively, with more GATA 4 expression in plasma than in nuclei. The expression of AFP was in plasma in carcinoma and adjacent tissue. GATA 4 and AFP expressions in carcinoma and adjacent tissue were significantly correlated and were significantly different. The expression of GATA 4 in carcinoma was higher than that in adjacent tissue, also higher in poorly differentiated carcinomas than in well differentiated ones. Conclusion; The expression of GATA4 and AFP can be detected both in hepatocarcinoma and adjacent tissues, with GATA4 being more sensitive. The transcription activity of GATA4 is closely corelated to the differentiation of hepatocarcinoma.
