摘要 目的:探讨去甲二氢愈创木酸(nordihydroguaiaretic acid,NDGA)对乳腺癌细胞MCF7增殖和凋亡的生物学特性的影响。方法:MTT法绘制细胞生长曲线及检测细胞存活率、倒置相差显微镜和扫描电镜观察细胞凋亡、流式细胞仪检测细胞周期和凋亡率。结果:NDGA对乳腺癌细胞MCF7生长增殖的抑制作用呈剂量和时间依赖效应,阻滞细胞周期于S期,诱导细胞凋亡形成,使细胞表面超微结构发生改变。结论:NDGA能够抑制乳腺癌MCF7细胞增殖,诱导细胞凋亡。 Objective: To explore the effects of nordihydroguaiaretic acid (NDGA) on apoptosis and proliferation of breast cancer MCF-7 cells. Methods: The cells growth curve and survival rate were observed by means of MTT asssy. Morphological variation was investigated by invert phase-contrast microscopy and scanning electron microscopy. Cell cycle analysis and apoptosis rate were evaluated by flow cytometry. Results: NDGA at various concentrations inhibited the growth of MCF-7 cells in a time-and dose-dependent manner, blocked the cell cycle in S phase, induced cells apoptosis and changed the morphology of MCF-7 cells. Conclusion: NDGA can inhibit the proliferation and induce the apoptosis of MCF-7 cells.
