八棱丝瓜蛋白2对B16细胞的诱导分化作用 被引量:2

摘要 目的测定八棱丝瓜蛋白2(LF-2)对小鼠黑色素瘤B16细胞的诱导分化作用。方法以MTT法测定LF-2对B16细胞的增殖抑制作用,以形态学、黑色素含量及细胞周期变化为指标,观察LF-2诱导B16细胞分化的作用。结果LF-2对B16细胞具有明显的增殖抑制作用,并使B16细胞停滞于G1期;细胞生长缓慢,平铺不重叠,甚至形成网状结构,呈正常上皮样细胞分化,细胞质内细胞器丰富,黑色素小体明显增多;黑色素含量显著增加。结论LF-2对B16细胞具有明显的诱导分化作用。 Objective To investigate the inducing differentiation effect of luffaculin-2 on B16 mouse melanoma ceils. Methods MTT assay was used to evaluate the inhibitory effect of luffaculin-2 on proliferation of B16 cells. Morphological changes, melanin content and distribution of cell cycle were also used to estimate the differentiation-inducing activity of luffaculin-2 in B16 ceils. Results Luffaculin-2 significantly inhibited the proliferation of B16 cells. Luffaculin-2 interrupted the transition of cell cycle from G1 phase to S phase. With luffaculin-2 treatment, B16 ceils were differentiated into mature epithelia-like ceils, grew slowly in monolayer and even formed reticulation, enriched cell organelles in cytoplasm, increased the number of melanosome obviously, and enhanced the melanin content markedly. Conclusion Luffaculin-2 can significantly induce differentiation of B16 cells.
