Expression of β-catenin and Cyclin D_1 in Laryngeal Squamous Cell Carcinoma and its Clinical Significance

摘要 目的探讨β-连环蛋白、细胞周期蛋白D1(Cyclin D1)在喉鳞癌的表达及其意义.方法采用免疫组织化学SP法检测58例喉鳞癌及10例正常喉上皮组织β-连环蛋白及Cyclin D1的表达.结果10例正常喉上皮β-连环蛋白表达位于细胞膜,细胞质未见表达;Cyclin D1均呈阴性表达.58例喉鳞癌组织中,31例β-连环蛋白细胞质表达,16例细胞核表达;34例Cyclin D1细胞核表达.pN+的β-连环蛋白细胞质表达阳性率明显高于pN0(P<0.05),T3~T4喉癌病例中β-连环蛋白细胞质表达阳性率明显高于T1~T2喉癌病例(P<0.05);β-连环蛋白细胞质阳性表达与喉鳞癌部位、分化程度无关(P>0.05).Cyclin D1阳性表达仅与喉癌浸润程度相关(P<0.05),而与喉癌发生部位、分化程度、淋巴结转移无关(P>0.05).31例β-连环蛋白细胞质表达喉癌组织中23例Cyclin D1阳性表达,明显高于β-连环蛋白细胞膜表达者.结论喉鳞癌中β-连环蛋白表达可作为评估喉鳞癌恶性程度的重要生物学指标. Objective To study the expression pattern of i3-catenin and cyclin D1 in laryngeal squamous cell carcinoma(LSCC) tissue, and discuss its clinical and pathological significance. Methods By using immunohistochemical staining(SP method), the expression of β-catenin and cyclin D1 were detected in 10 laryngeal normal membrane and 58 LSCC. Results In laryngeal normal tissue, β-catenin showed high intense expression at the membrane and negative expression at the cytoplasm and nucleus. Cyclin D1 showed negative expression at the nucleus. In contrast to the normal tissue, β-catenin was expressed variously in the cytoplasm in carcinoma but less, or even lost expression at the membrane in cancer samples. In 16 cases, β-catenin could be detected in the nucleus, the positive expression of cyclin D1 was mainly confined to the nuclei(34/58). Moreover, the positive rate of β-catenin cyclin D1 expression in cytoplasm was significantly higher in those patients with lymph node metastasis and T3-4 staging than patients with no lymph node metastasis and T1-2(P 〈 0.05). But had no correlation with the tumor differentiation, location(P 〉0.05). The positive expression of cyclinD1 had only a correlation with invasive depth, but not correlated with the tumor differentiation, location, lymph node metastasis. In LSCC, the aberrant expression of β-catenin in cytoplasm and cyclin D1 in nucleus was positively related to the expression of cyclin DI(P〈0.05), Oonclusion β-catenin and cyclin D1 expression could be used as an important biological indices for evaluating the malignancy of LSCC.
