摘要
目的探讨秋水仙碱(Colchicine)在心肌肥厚发生、发展中的作用及对心肌、血浆中血管紧张素Ⅱ(AngiotensinⅡ,AngⅡ)水平和心肌中转化生长因子-β1(TrnsforminGrowthFactor-beta1,TGF-β1)基因表达的影响。方法用腹主动脉缩窄的方法建立心肌肥厚大鼠模型,应用放射免疫分析法测定体内AngⅡ水平,RT-PCR法半定量TGF-β1的表达变化。结果发现腹主动脉缩窄2周、4周后大鼠心脏质量指数,心肌、血浆中AngⅡ水平及心肌TGF-β1的表达均明显升高,经过Colchicine2周、4周干预后可显著降低心脏质量指数,AngⅡ及TGF-β1的表达水平。结论Colchicine能有效抑制心肌肥厚发生、发展,同时对心肌肥厚相关因子表达有明显的下调作用。
objective To investigate the effect of colchicine on cardiac hypertrophy and circulating and ventricular Angiotensin Ⅱ (Ang Ⅱ ) and ventricular transforming growth factor-beta 1 (TGF-β1). Methods Circulating and cardiac muscle Ang Ⅱas measured by radioimmunoassay, ventricular TGF-β1 was examined by RT-PCR after abdominal aorta constriction (AC). Results The levels of Ang Ⅱ , TGF-β1 and ratios of overall or left heart weight to body weight were significantly increased after AC two weeks or four weeks and significantly decreased after treatment with colchicine. Conclusion These results suggest that colchicine can inhibit the genesis of cardiac hypertrophy and down regulated the expression of correlated factors.
