非小细胞肺癌组织中COX-2和VEGF的表达及其与肿瘤血管生成的研究 被引量:5

Expressions of COX-2 and VEGF in non-small cell lung cancer and the relationships between them and tumor angiogenesis

摘要 目的探讨环氧化酶-2(COX-2)和血管内皮生长因子(VEGF)在非小细胞肺癌(NSCLC)组织中的表达及其与肿瘤生物学特征和肿瘤血管生成的关系。方法免疫组化法检测NSCLC组织COX-2和VEGF的表达,并测定肿瘤微血管密度(MVD)。结果⑴NSCLC组织中COX-2及VEGF阳性表达率分别为55.5%,66.7%,显著高于正常肺组织(P均<0.05);⑵有淋巴结转移组COX-2阳性表达率高于无淋巴结转移组;病理分级中Ⅱ-Ⅲ级组COX-2阳性表达率高于Ⅰ级组;肺腺癌组COX-2的阳性表达率高于肺鳞癌组,差异均有显著性(P均<0.05);⑶T分期中T3-4期组VEGF的阳性表达率高于T1-2期组,有淋巴结转移组VEGF阳性表达率高于无淋巴结转移组;病理分级中Ⅱ-Ⅲ级组VEGF阳性表达率高于Ⅰ级组;临床分期中Ⅲ-Ⅳ期VEGF的阳性表达率高于Ⅰ-Ⅱ期,差异均有显著性(P均<0.05);⑷COX-2与VEGF、MVD,VEGF与MVD的表达之间均呈正相关(r=0.696、0.673、0.571,P均<0.01)。结论COX-2和VEGF的增强表达与非小细胞肺癌的浸润性发展和肿瘤血管生成有密切关系,VEGF可能参与COX-2对肿瘤血管生成的促进作用。 Objective To investigate the expressions of Cycloxygenase-2(COX-2) , vascular endothelial growth factor(VEGF) in non-small cell lung cancer (NSCLC) to explore the relationships between them and the tumor biological characteristics,tumor angiogenesis in NSCLC. Methods The expressions of COX-2 ,VEGF and the microvessel density(MVD) were detected by immunohistochemical staining. Results The positive rates of COX-2 and VEGF expression in NSCLC tissue were 55.5% and 66. 7% respectively, with significantly higher than those of normal lung tissue( all P〈0. 05 ). The positive rate of COX-2 expression was significantly higher in the tissue of NSCLC patients with lymph node metastasis than in those without lymph node metastasis, and higher in patients of pathological grade Ⅱ- Ⅲ than in those of grade Ⅰ , and higher in patients of adenocarcinoma carcinoma than in those of squamous cell carcinoma ( all P〈0.05 ). The positive rate of VEGF expression was significantly higher in patients of T stage T3-4 than in those of stage T1 -2, and higher in patients with lymph node metastasis than in those without lymph node metastasis, and higher in patients of pathological grade Ⅱ-Ⅲ than in those of grade Ⅰ, and higher in patients of clinical stage Ⅲ-Ⅳ than in those of stage Ⅰ-Ⅱ ( all P 〈0. 05). There was statistically significant correlation between expression of COX-2 and VEGF, COX-2 and MVD, VEGF and MVD, respectively( all P 〈0. 01 ). Conclusion The high expressions of COX-2 and VEGF in NSCLC tissues may be related to tumor invasion and angiogenesis.
