抗组胺药抑制长波紫外线辐射诱导HaCaT细胞生成IL-6

Antihistamine Reagents Inhibites the Effect upon the Release of IL-6 in HaCaT Cell Line under Ultraviolet A Radiation

摘要 目的观察长波紫外线辐射对体外培养的HaCaT细胞产生IL-6的影响及抗组胺药的作用。方法用不同剂量长波紫外线辐射培养的HaCaT细胞,加入不同浓度的异丙嗪及西咪替丁(分别为0,10-8,10-7,10-6,10-5mol/L),在不同时点收集上清液,ELISA检测IL-6浓度。结果与未辐射组相比,HaCaT细胞经50,100,200及500 KJ/m2剂量的长波紫外线辐射后12 h开始IL-6分泌均显著增加(P<0.05);与未加抗组胺药组相比,HaCaT细胞经200 KJ/m2辐射加入异丙嗪或(和)西咪替丁后24 h均可显著抑制IL-6的产生(P<0.05),且异丙嗪加西咪替丁组及异丙嗪组比西咪替丁组更显著(P<0.05)。结论长波紫外线辐射可刺激HaCaT细胞产生IL-6,抗组胺药可抑制这种作用。 Objective To study the effect on the release of IL-6 in HaCaT cell line under ultraviolet A radiation and the effect of antihistamine reagents upon it. Methods HaCaT keratinocytes were incubated without or with promethazine or (and) cimetidine (eoncentration 10^-8, 10^ -7,10^ -6 and 10^-5 mol/L) under various ultraviolet A treatment modalities in vitro, supernatants of the cell cultures were recovered and IL-6 was detected by ELISA. Results Compared with no radiation ,an increase of IL-6 in vitro upon ultraviolet A radiation from HaCaT keratinocytes was observed from 12h for 50,100,200 and 500 KJ/m^2 respectively( P 〈 0.05). The production of IL-6 induced by ultraviolet A radiation from HaCaT keratinocytes was inhibited after promethazine or (and) cimetidine were added into cultures compared with no antihistamine reagents (P 〈 0.05 ). Conclusion These results suggest that ultraviolet A radiation can induce the release of IL-6 in HaCaT cell line and antihistamine reagents can inhibit the effect.
