摘要
目的:研究凋亡相关基因Fas和FasL在鼻咽癌患者诱导化疗前后外周血单个核细胞中表达水平的变化,探讨其在鼻咽癌发生、发展中的作用及意义。方法:采用流式细胞仪对38例鼻咽癌患者诱导化疗前后及15例正常健康对照组外周血单个核细胞中的Fas和FasL表达进行检测,并结合临床资料进行分析。结果: ( 1 )鼻咽癌患者外周血单个核细胞中Fas、FasL的表达明显高于正常健康对照组(P<0.05);Ⅲ+Ⅳ期患者的表达明显高于Ⅱ期患者(P<0.05)。(2)T3+4期患者外周血单个核细胞Fas表达明显高于T1+2期患者(P<0.05),而其FasL的表达稍高于T1+2期患者(P>0.05)。(3)N1 期患者的Fas表达明显高于N0 期患者(P<0.05),而其FasL的表达稍高于N0 期患者(P>0.05);N2+3期患者的Fas表达明显高于N1期(P<0 .05),而其FasL表达稍高于N0 期和N1 期(P>0.05)。(4)鼻咽癌患者经诱导化疗后,其Fas表达明显增高(P<0.05),FasL表达稍有增高(P>0.05)。结论:外周血单个核细胞Fas、FasL表达水平与鼻咽癌发生、增殖、局部转移有关,对其进行检测可作为鼻咽癌的诊断及病期判断的辅助指标。
Objective:To study the expression and significance of relevant apoptosis genes, Fas and FasL, on peripheral blood mononuclear cells (PBMC) of patients with nasopharyngeal carcinoma (NPC) before and after the induction chemotherapy, and to investigate their function and clinical value in occurrence, development and treatment of NPC. Methods:Expression of Fas and FasL on PBMC of 38 patients with NPC before and after the induction chemotherapy and 15 normal healthy controls were tested with flow cytometry, and analysed with clinical information. Results:①The expression of Fas and FasL on PBMC in NPC patients was much higher than that in normal healthy controls ( P 〈0. 05 ). The expression of Fas and FasL on PBMC in NPC patients of stage Ⅲ + Ⅳ were much higher than that in stage Ⅱ ( P 〈 0. 05 ). ②The expression of Fas on PBMC in NPC patients of Stage T3 ,4 was much higher than that in stage T1 ,2 ( P 〈 0. 05 ) , but there was no obvious change in expression of FasL ( P 〉 0. 05 ). ③ The expression of Fas on PBMC in NPC patients of stage N1 was much higher than that in stage NO ( P 〈 0. 05 ), there was no obvious change in expression of FasL ( P 〉 0. 05 ). The expression of Fas on PBMC in NPC patients of stage N2+3 was much higher than that in N1 (P 〈0. 05) , but there was no obvious change in expression of FasL ( P 〉 0. 05 ). ④For NPC patients received induction chemotherapy, the expression of Fas increased obviously compared with it before the chemotherapy ( P 〈 0. 05) , there was no obvious change in expression of FasL ( P 〉 0. 05 ). Conclusion :The expression of Fas and FasL on PBMC shows certain significance for occurrence, multiplication and local metastasis of NPC. It can also help diagnose of NPC and judge its stage.
