摘要 目的研究溶血磷脂酸(LPA)受体在人卵巢癌细胞系中基因和蛋白的表达及意义。方法采用半定量RT-PCR、免疫细胞化学链霉菌抗生物素蛋白·过氧化物酶连接(SP)法和Westernblotting检测LPA1、LPA2和LPA3mRNA以及LPA2和LPA3蛋白在人卵巢癌细胞系3AO·SKOV3·OVCAR3中的表达水平。结果(1)RT-PCR结果显示,LPA1、LPA2和LPA3mRNA在人卵巢癌细胞系中均有不同程度的表达;(2)免疫细胞化学和Westernblotting结果显示,LPA2和LPA3蛋白在人卵巢癌细胞系中均有表达。结论LPA1、LPA2和LPA3在人卵巢癌细胞系中广泛表达,LPA2和LPA3可能参与了卵巢癌的发生发展过程。 Objective To investigate the expressions of lysophosphatidic acid (LPA) receptors at both mRNA and protein levels and their biological significance in different human ovarian cancer cell lines. Methods Reverse transcription-polymerase chain reaction (RT-PCR), streptovidin peroxidase-conjugated (SP) immunohistochemical assay and Western blotting were employed to measure the expression levels of LPA1, LPA2 and LPA3 mRNA and LPA2 and LPA3 protein in cultured human ovarian cancer cell lines 3AO. SKOV3, and OVCAR3. Results All the ovarian cancer cell lines expressed LPA1, LPA2 and LPA3 mRNAs and LPA2 and LPA3 proteins. Conclusion LPA1, LPA2 and LPA3 expressions occur extensively in human epithelial ovarian cancer cell lines, and LPA2 and LPA3 may be involved in the development and progression of human ovarian cancer.
