酒精性股骨头缺血坏死大鼠血浆内皮素与一氧化氮的变化 被引量:7

摘要 目的:观察酒精性股骨头缺血坏死模型大鼠的内皮素与一氧化氮含量的变化,分析其在股骨头缺血性坏死发病中的作用。方法:实验于2003-07/2004-04在福建中医学院实验动物中心进行。SD大鼠30只,随机分为3组,采用酒精灌胃法造模,高剂量组给予56%白酒8mL/(kg·d)灌胃;低剂量组给予56%白酒6m L/(kg·d)灌胃;对照组给予生理盐水灌胃,3组动物于20周麻醉处死取材,进行一氧化氮和内皮素含量的测定。结果:实验大鼠30只均进入结果分析。①高剂量组与低剂量组动物的血浆内皮素含量比对照组明显升高,尤其高剂量组升高最为显著[对照组:(110.46±26.01)ng/L,高剂量组(179.80±37.37)ng/L,P<0.01;低剂量组(149.01±32.70)ng/L,P<0.05]。②高剂量组与低剂量组动物的血清一氧化氮含量比对照组降低,高剂量组与对照组有显著性差异[对照组:(19.00±4.89)μmol/L,高剂量组(13.71±4.24)μm ol/L,P<0.05;低剂量组(15.35±4.30)μmol/L,P<0.05]。结论:酒精中毒导致内皮素含量升高和一氧化氮含量降低,引起股骨头微循环障碍,最终导致其缺血性坏死。 AIM: To observe the changes of contents of endothelin and nitric oxide in rats models of alcoholic avascular necrosis of the femoral head, and analyze the roles in the attack of avascular necrosis of the femoral head. METHODS: The experiment was carried out in the experimental animal center of Fujian College of Traditional Chinese Medicine between July 2003 and April 2004. Thirty SD rats were randomly divided into 3 groups, and they were made into models by means of gastric perfusion of alcohol. Rats in the high and low dosage groups were given 56% white wine for 8 and 6 mL/kg per day respectively, and those in the control group were given saline. All the rats in the 3 groups were killed under anesthesia at 20 weeks to determine the contents of endothelin and nitric oxide. RESULTS: All the 30 rats were involved in the analysis of results. ① Compared with the control group, the content of endothelin in plasma were obviously increased in the high and low dosage groups, and more significantly in the high dosage group [control group: (110.46±26.01) ng/L, high dosage group: (179.80±37.37) ng/L, P 〈 0.01; low dosage group: (149.01±32.70) ng/L, P〈0.05]. ① Compared with the control group, the content of nitric oxide in plasma were decreased in the high and low dosage groups, and there were significant difference between the high dosage group and control group [control group: (19.00±4.89)μmol/L, high dosage group: (13.71±4.24)μmol/L, P 〈 0.05; low dosage group: (15.35±4.30)μmol/L, P 〈 0.05]. CONCLUSION: Alcoholism can cause the increase of the content of endothelin and the decrease of the content of nitric oxide, and then leads to the disorder of microcirculation of femoral head, final results in its avascular necrosis.
