摘要 目的观察醛固酮(ALDO)对血管外膜肾上腺髓质素(ADM)产生和分泌的影响以及ADM mRNA表达水平,以探讨ADM和ALDO之间相互联系。方法用放射免疫法测定ALDO刺激后ADM产生和分泌水平,用3H-THR掺入反映血管外膜增殖,半定量的RT-PCR测定ADM基因表达。结果ALDO呈浓度依赖的刺激大鼠血管外膜ADM分泌和mRNA表达,ADM抑制ALDO诱导的外膜增殖。ALDO活化外膜成纤维细胞丝裂原活化蛋白激酶(MAPKs),明显增加其活性,该效应可被ADM抑制。结论ALDO促进血管外膜ADM的生成,而生成的ADM拮抗AL-DO诱导的血管外膜增殖。 Objective: To examine the effect of aldosterone on adrenomedullin(ADM) production and secretion and its mRNA expression in rat aortic adventitia to study the paracrine/autocrine interaction between endogenous ADM and aldosterone. Method: ADM produced and secreted from adventitia stimulated by aldosterone was detected by radioimmunoassay.Proliferation in adventitia cells was evaluated by the level of 3H-thymine incorporation.And prepro ADM gene expression was measured by semi-quantitative reverse transcriptase polymerase chain reaction. Results: Adventitial ADM secretion and mRNA expression stimulated by aldosterone were concentration-dependent and it was an inhibitive effect of ADM on aldosterone-induced proliferation. ADM inhibited the activity of mitogen activated protein kinases(MAPKs) stimulated by aldosterone.Conclusion: Aldosterone stimulates adventitia to produce and secrete ADM, which in turn antagonizes the aldosterone-induced proliferation in adventitia.
