摘要
以低次烟叶为材料,研究了烟叶蛋白质最佳提取工艺。采用L9(34)正交试验设计研究了烟叶磨浆 工艺。结果表明,磨浆工艺的最佳工艺参数为固液比(烟叶:水)1:17,提取温度60℃,提取液pH 8.0,磨浆2次。 采用响应曲面分析法研究了烟叶蛋白碱溶工艺,得到其最佳工艺参数为温度60℃,pH 8.0,时间60 min,搅拌条件 下碱溶3次。烟叶蛋白酸沉操作中,在pH 3.0,4℃下静置8 h效果最佳,最终烟叶蛋白提取率为76.62%。
The optimum extracting conditions of protein in discarded tobacco leaf was studied. The results showed that the optimum technology conditions for tobacco leaf grinding were as follows :the ratio of 1 (tobacco leaf) ; 17(water),temperature 60 C ,pH 8.0 and grinding twice with orthogonal experiment design methods L9(34). The optimum extracting conditions of protein were 60 C ,pH 8.0,60 min and extracting three times with agitation by Response Surface Methodology. And the optimum conditions of depositing protein with acid were pH 3.0,4 C for 8 h. The extraction rate reached 76.62% under the optimal conditions.
