摘要 目的:研究鼻咽癌组织中P73基因的表达及其临床意义.方法:运用逆转录聚合酶链反应技术(RT-PCR)及单链构象多态性分析技术(SSCP)检测P73mRNA的表达及P73基因的突变情况.运用免疫组化SABC技术检测P73蛋白的表达情况.结果:47例鼻咽癌组织中29例P73mRNA表达阳性,15例鼻咽粘膜慢性炎症组织中3例P73mRNA表达阳性.鼻咽癌组织中P73mRNA的表达与鼻咽粘膜慢性炎症组织相比,其差异具有显著性(P<0.05).鼻咽癌组织中P73mRNA的表达与TNM分期存在密切关系,Ⅲ~Ⅳ期患者P73mRNA的阳性表达率较Ⅰ~Ⅱ期高(P<0.05).P73基因在所检测的标本中未发现有突变.免疫组化技术检测证实鼻咽癌中存在P73蛋白的过表达(P<0.05).结论:鼻咽癌组织中存在P73基因的过表达现象,P73基因的异常表达与鼻咽癌的发生发展存在一定的关系. Objective: To investigate the expression and significance of P73 gene in nasopharyngeal carcinoma. Methods: The expression of P73 mRNA was detected by reverse transcriptase polymerase chain reaction(RT-PCR). We performed analysis of mutation of P73 gene by PCR-single strand conformation polymorphism (SSCP) analysis, and P73 protein was examined by using immunochistochemical SABC staining method. Results: Overexpression of P73 mRNA was found in 29 nasopharyngeal carcinoma tissues among 47 cases, in 3 chronic nasopharynxitis among 15 cases. The positive expression rate of P73 mRNA in nasopharyngeal carcinoma tissues was significantly higher than that in chronic nasopharynxitis tissues (P〈 0. 05). The positive expression rate of P73 mRNA in nasopharyngeal carcinoma tissues of stage Ⅲ - Ⅳ group was significantly higher than that of stage Ⅰ -Ⅱ group. No mutation of P73 gene was found in all detected cases. The strong expression of P73 gene in nasopharyngeal carcinoma tissues was proved by immunochistochemical staining method. Conclusion: There is an overexpression of P73 gene in nasopharyngeal carcinoma, and their expression is relationship counected with oncogenesis and development of nasopharyngeal carcinoma.
