摘要
目的探讨中风康对局灶性脑缺血大鼠脑组织水通道蛋白mRNA(AQP-4m RNA)表达的影响。方法采用线栓法复制局灶性脑缺血大鼠模型,于脑缺血12,24,48,72,96h观察模型大鼠AQP-4m RNA表达的动态变化,并在脑缺血48h观察中风康对AQP-4m RNA表达及脑水肿的影响。结果与假手术组比较,局灶性脑缺血12h,模型组脑组织AQP-4m RNA表达开始增加,至72h达到高峰(P<0.01),与48h模型组比较,各治疗组脑组织AQP-4mRNA表达和脑组织含水量均有不同程度降低(P<0.05或P<0.01)。结论中风康可降低脑梗塞大鼠脑组织AQP-4mRNA表达,减轻脑水肿。
Objective To observe the effect of Zhongfengkang on the expression of Aquaporin 4 mRNA (AQP-4mRNA) in brain of focal cerebral ischemia rats. Methods The rat model of focal cerebral ischemia was established by inserting nylon thread. The dynamic changes of AQP-4 mRNA expression in the model group at 12^th 24^th, 48^th 72^nd and 96^th hour after cerebral ischemia were examined and the effect of Zhongfengkang on AQP-4 mRNA expression and cerebral edema at 48^th hour after cerebral ischemia was observed. Results At 12^th hour after cerebral ischemia, the expression of AQP-4mRNA in the model group increased compared with the sham-operation group, and it arrived to the peak at 72^th hour (P〈0.01) ; Compared with model group at 48^th, the expression of AQP-4 mRNA and the brain edema in each treatment groups were decreased (P〈0.05 or P〈0.01) . Conclusion By decreasing the expression of AQP-4 mRNA in brain of focal cerebral is- chemia rats, Zhongfengkang could protect blood brain barrier and reduce cerebral edema.
