摘要
目的探讨TSP1/TGFβ及相关因子与垂体瘤血管生成、侵袭性的关系及意义。方法取46例垂体瘤手术标本,应用半定量RT-PCR检测TSP1及其受体CD36与TGFβ的表达,采用免疫组化检测TSP1/TGFβ及MVD,并分析TSP1/TGFβ表达与侵袭性、MVD等因素之间的关系。结果侵袭组TSP1表达明显低于与非侵袭组,而TGFβ表达则相反;两组间MVD、CD36的表达无统计学意义;TSP1表达与侵袭性呈负相关,TGFβ与侵袭性及MVD呈正相关。结论TSP1/TGFβ在垂体瘤发生发展中起重要作用,并影响血管生成及侵袭性,有望成为治疗垂体瘤的新靶点。
Objective To explore the expression of TSP1, TGFβ, CD36 in pituitary adenomas and investigate the relationship between the expression and invasion, microvessel density of adenomas. Methods The expression of TSP1, TGFβ, CD36 in 46 surgical specimens( invasive cases, 21 ;noninvasive cases,25) clinical pituitary adenomas was studied by means of semi-quantify RT-PCR and immunohistochemistry method,and the relationship between the expression and invasion of adenomas was explored . MVD was measured by immunohistochemical method with detecting CD34. Results As followed,a negative correlation was demonstrated between TSP1 (t = 4. 356 ;P = 0. 001 ) expression and the invasion of pituitary adenomas; The expression of TGFβ(t =3. 318;P =0. 005) and MVD (t = 1. 135;P = 0. 305) was increased in invasive cases than in noninvasive ones, thus it had a positive correlation with MVD; There was no correlations between expression of CD36 ( t = 2. 257 ;P = 0. 083 ) and the invasiveness and MVD of pituitary adenomas. Conclusions TSP1/TGFβ might play an important role in the development of pituitary adenomas,and TSP1/TGFβregulates invasiveness and angiogenesis. Thus, TSP1/TGFβ might be a new therapy target in pituitary adenomas.
