摘要
【目的】探讨细胞凋亡在矽肺肺纤维化过程中的作用。【方法】随机将Wistar大鼠分为对照组和染矽尘组。采用暴露式气管内一次注入二氧化硅粉尘悬液建立大鼠矽肺模型。模型建立后,分6个时间点(1、3、7、14、21、28 d),每组分别取8只大鼠将其处死取肺组织,用TUNEL法观察大鼠肺组织不同时间点细胞凋亡的情况,并采用Image-Pro Plus Version 4.5 for windowsTM图像分析计算积分光密度。【结果】大鼠矽肺模型肺组织凋亡阳性细胞在染矽尘后的1、3、7、14、21、28 d时,显著高于对照组,有显著性差异(P<0.05)【结论】染矽尘大鼠肺组织细胞凋亡与矽肺纤维化的发生发展有关。
[Objective] To study the role of apoptotic cells in the pulmonary fibrosis in silicosis. [Methods] Wistar rats were randomized into two groups: control group and experimental group. After the rats model of silicosis was established by intratracheal infusion of silica dust suspension, eight rats from each group were sacrificed at 1, 3, 7, 14, 21, and 28 d. The expression of the apoptotic cells induced by silica dust was determined by tissuechip microarray and TdT-mediated dUTP nick end-labeling method. The integral optical density of positive cells were quantitatively analyzed using Image-Pro Plus Version 4.5 for windows. [Results] The expression of apoptotic cells in the lung tissue of the model rats at 1, 3, 7, 14, 21 and 28 d were significantly more than those in the control group, and peaked at 7 - 14 d after exposure to silica. [Conclusion] The expression of apoptotic cells in the lung pulmonary tissue play a role in the pulmonary fibrosis in silicosis.
