摘要
【目的】探讨染矽尘大鼠肺组织Ⅰ、Ⅲ型胶原表达变化的意义。【方法】将96只大鼠随机分为对照组、染矽尘组,采用一次性气管内注入矽尘法建立矽肺动物模型,染尘后按不同时间点取大鼠肺组织,用天狼猩红染色法结合偏振光显微镜观察Ⅰ、Ⅲ型胶原的表达,Image-Pro Plus图像分析系统进行定量分析。【结果】与对照组比较,染矽尘大鼠肺组织Ⅰ、Ⅲ型胶原表达增强,各时间点胶原面积百分比均有不同程度的增加,Ⅰ型胶原在染矽尘后7 d、Ⅲ型胶原在染矽尘后3 d开始增加,两种胶原在28 d时增加幅度最大。【结论】矽尘诱导染矽尘大鼠肺组织Ⅰ、Ⅲ型胶原的增生,且这两种胶原的增生具有时相性。
[Objective] To study the temporal expression of type Ⅰ、Ⅲ collagen in lung tissue of silica-treated rat. [Methods] The 96 experimental rats were randomly divided into control and silica group, 1ml silica (50 g/l) was injected intratracheally in rats of silica group. Samples were collected on the 1, 3, 7, 14, 21, 28 d after injected silica. Expression of Ⅰ、Ⅲ collagen were stained with sirius red and detected by polarized light microscopy and Image-Pro Plus. [Results] Compared with control group, at different time points type Ⅰ、Ⅲ collagen expression of silica group increased at different levels. Type Ⅰ collagen begins to increase on the 7 d, as well as type Ⅲ collagen on the 3 rd day. The two types of collagen increase the most markedly on the 28 d. [Conclusion] Silica dioxide induce the increase of type Ⅰ、Ⅲ collagen production in lung tissue, and their stages are different.
