神经生长因子对Aβ_(1-40)诱导海马神经元周期蛋白异常表达的影响

摘要 为了探讨神经生长因子(NGF)对β-淀粉样蛋白诱导海马神经元周期蛋白表达的影响。本研究取新生3d内大鼠海马细胞行体外培养7d,分别在加入Aβ1-40前后加入NGF和等体积培养液,采用β-tubulin、ChAT免疫组化、cyclin免疫荧光进行检测。β-tubulin免疫组化证实所培养的为特异性结构完整的海马神经元;cyclin免疫荧光检测、ChAT免疫组化显示Aβ1-40导致cy-clin A、cyclin B1水平显著增高及ChAT免疫阳性细胞明显减少;而给予NGF可在不同程度上降低cyclin A、cyclin B1蛋白水平,增加ChAT免疫阳性细胞。结果提示,NGF能有效地阻止和轻度地修复Aβ1-40诱导的海马细胞损伤,对Aβ引起的海马神经元内异常表达细胞周期蛋白有一定的抑制作用。 To observe the effects of nerve growth factor(NGF) on the aberrant neuronal expression of mitotic protein in hippocampal neurons of new-born SD rats with β-amyloid(Aβ1-40). Hippocampal neurons obtained from new-born rats within 3 d were cultured for 7 d, then Aβ1-40(10μg/ml ) was added before and after addition of NGF(10 ng/ml ) and equal amount of culture fluid separately. β-tubulin and ChAT immunohistochemistry were used to determine the integrity of hippocampal neurons and ChAT-positive neurons. The expression of mitotic protein was evaluated with immunocytochemistry by cyclin A and cyclin B1 respectively. β-tubulin immunohistochemistry demonstrafed the integrity of hippocampal cells: ChAT and cyclin immunoreaction showed that the number of ChAT-positive neurons were decreased significantly, and the expression of cyclin A and cyclin B1 protein has hoisted in Aβ1-40 group. NGF, however, could improve the above-mentioned indexes in yawing degrees. These results demonstrate that the efficacy of NGF may be partly results from its improving the expression of cell cycle related protein of hippoeampal neurons induced by Aβ1-40.
