摘要 比较1B x 14及其它已知HMW-GS基因的启动子和编码区,根据其不同点设计出1B x 14基因特异扩增引物.以8种已知HMW-G S组成的小麦DNA为模板进行PCR扩增,结果表明:具有1Bx14亚基的品种扩增出1条400 bp左右特异条带.结合该特异标记和已报道的1Dx5特异标记对2个F2杂交群体进行检测,从184个F2单株中筛选出111个同时含有1B x 14和1D x 5基因的单株.该研究结果可为种质鉴定和亚基整合育种提供参考. Based on the known HM-W-GS1Bx14 promoter and coding region,the specific primers for 1Bx14 were designed and synthesized. Using the genomic DNA from 8 wheat cultivars,a 400 bp or so fragment was amplified in 4 materials carrying the 1Bx14 subunit,while others didn't. The results showed that the primers were specific for HMW-GS1Bx14 gene. F2 generation population was detected with 1Bx14 and reported 1Dx5 marker. 111 plants with 1Bx14 and 1Dx5 gene were selected from 184 plants. This system could be applied in marker associate breeding and germplasm identity.
