肾缺血预处理对兔未成熟心肌Bcl-2,Bax,Fas蛋白表达的影响 被引量:5

摘要 目的:探讨肾缺血预处理(R IP)对兔未成熟心肌细胞凋亡和Bcl-2,Bax,Fas蛋白表达的影响.方法:24只幼兔采用Langendorff离体心脏灌注模型,随机分为正常对照组(C)、心脏缺血/再灌注组(I/R)和肾缺血预处理组(RIP),每组8只,测定各组未成熟心肌细胞凋亡和Bcl-2,Bax,Fas蛋白表达.结果:RIP组与I/R组比较,心肌细胞凋亡率明显减少[(4.19±1.13)%vs(12.30±2.10)%,P<0.01],DNA片段梯光密度明显减少(57 350±1765vs135 200±3370,P<0.01),Bcl-2表达增多(33 525±1026vs12 385±860,P<0.01),Bax(8640±768vs32 472±1128,P<0.01)和Fas(8936±912vs32 100±1260,P<0.01)表达明显减少.结论:RIP可减少缺血再灌注后兔未成熟心肌细胞凋亡和影响Bcl-2,Bax,Fas蛋白的表达. AIM: To investigate the effects of renal ische- mic preconditioning (RIP) on apoptosis and expressions of Bcl-2, Bax and Fas proteins in immature myocardial cells in rabbits. METHODS: Isolated working rabbit heart model was used in this study and 24 immature rabbits were ran- domly divided into control group, ischemia/reperfusion ( I/ R) group and RIP group. The TUNEL, nucleosomal ladder of DNA fragments and the expression of Bcl-2, Bax and Fas proteins were detected. RESULTS: Compared with that in I/R group, TUNEL cell apoptosis was lower [ (4.19 ±1.13)% vs (12.30±2.10)%, P〈0.01], ladder of DNA fragments was lower (57350 ±1765 vs 135 200±3370, P〈 0.01), Bcl-2 expression was higher (33 525 ± 1026 vs 12 385 ± 860, P 〈 0. 01 ), Bax expression was lower ( 8640 ± 768 vs 32 472 ± 1128, P 〈0. 01 ) and Fas expression was lower (8936 ± 912 vs 32 100 ± 1260, P 〈0.01 ) in RiP group. CONCLUSION: RIP decreases immature myocardial cell apoptosis and affects the expressions of Bcl-2, Bax and Fas proteins.
