摘要
目的:研究抗纤I号对肝星状细胞(HSC)的影响及抗肝纤维化的机制.方法:用RT-PCR方法检测转化生长因子β1(TGF-β1)和血小板衍生生长因子(PDGF)mRNA在HSC的表达,用W estern B lot检测TGF-β1和PDGF蛋白的表达.同时,应用MTT法观察HSC的生长曲线.结果:抗纤I号复方药物血清作用于HSC后,与对照组和秋水仙碱组相比HSC增殖明显被抑制(P<0.05),同时TGF-β1和PDGF蛋白及mRNA的表达明显下降(P<0.05).结论:抗纤I号复方药物血清能够抑制HSC增殖,下调TGF-β1和PDGF蛋白及mRNA的表达,这可能是该药抗肝纤维化的分子机制之一.
AIM: To investigate the effects of anti-fibrosis herbal compound on activated hepatic stellate cells and to explore its molecular mechanism in the treatment and prevention of hepatic fibrosis. METHODS: The transforming growth factor-β1 (TGF-β1) and platelet derived growth fac- tor (PDGF) mRNA were detected by RT-PCR method. The protein of TGF-β1 and PDGF was determined by Westem Blot analysis. The cell growth curve was also observed with MTT assay. RESULTS: After treated with 100 mL/L drug serum of anti-fibrosis herbal compound and 100 mL/L colchicine serum for 24 h, the mRNA and protein expression of TGF-β1 and hepatic stellate cells' PDGF decreased sharply compared with those in control group ( P 〈 0. 05). The mRNA and protein expression of TGF-β1 and PDGF in anti-fibrosis herbal compound group was lower than that in colchicine group ( P 〈 0. 05 ). MTT assay revealed that the proliferation of the activated hepatic stellate cells in colchicine group and anti-fibrosis herbal compound group reduced significantly compared with those in control group (P 〈 0. 05). CONCLUSION: Anti-fibrosis herbal compound can treat hepatic fibrosis by inhibiting the expression of TGF-β1 and PDGF of the activated hepatic stellate cells.
