辐射法降解壳聚糖及其抑菌性能的研究 被引量:15

STUDY ON RADIATION-INDUCED DEGRADATION OF CHITOSAN POWDER AND ITS ANTIBACTERIAL PROPERTIES

摘要 采用γ射线辐照降解壳聚糖,并利用凝胶渗透色谱和傅立叶红外对辐照所得产物进行分析。发现在100 kG y剂量时,就能有效降解壳聚糖,所得产物的分子量约为原来的四分之一。在50 kG y^300 kG y的剂量范围内,辐照对其结构和脱乙酰度几乎没有影响。针对一种革兰氏阳性菌(金黄色葡萄球菌,S t.aereus)和三种革兰氏阴性菌(大肠杆菌,E.co li;弧菌,V.angu illarum;气单胞菌,A.hydroph ila),试验了辐射降解壳聚糖低聚物的抑菌性能。发现在实验的分子量范围内,壳聚糖低聚物对革兰氏细菌均有良好的抑制作用。在壳聚糖质量分数为3×10-4时,抑菌率大于90%。 Low-molecular-weight chitosan (chit-oligomer) was prepared by irradiation of chitosan powder in the air and chit-oligomer's antibacterial properties were tested against different bacteria. Gel permeation chromatography measurement and FT-IR analysis indicate that radiation causes serious decrease of chitosan's molecular weight but without changing its chemical structure and degree of deacetylation up to 300 kGy. Antibacterial properties both for gram positive bacteria (St. aereus) and negative ones (E. coli, V. anguillarum, A. hydrophila) were studied by adding chitosan solution into the cultivated medium. Chit-oligomers showed good antibacterial effects against all the bacteria tested. The antibacterial efficiency was also dependent on the molecular weight of chit-oligomer or the irradiation dose.
