摘要
目的:研究头痛宁对大鼠血小板功能、耐缺氧能力的影响及镇痛机制。方法:取体重(180±20)gWistar大鼠200只,随机分为4组,分别灌胃给予生理盐水、头痛宁大、小剂量和麦角胺咖啡因片,测定用药后大鼠出凝血时间,ADP诱导的大鼠血栓性死亡的死亡率,寒凝血瘀大鼠血粘度和红细胞聚集指数及密闭缺氧存活时间。结果:头痛宁组大鼠用药后出凝血时间明显延长,ADP诱导的大鼠血栓性死亡的死亡率、寒凝血瘀大鼠血粘度和红细胞聚集指数均低于对照组,大鼠密闭缺氧存活时间长于对照组,且头痛宁高剂量组与麦角胺咖啡因组及头痛宁小剂量组比较,差异显著(P<0.05)。结论:头痛宁可明显延长大鼠出凝血时间,降低ADP诱导的大鼠血栓性死亡的死亡率,明显改善寒凝血瘀大鼠循环障碍,改善微循环,降低血粘度和红细胞聚集指数,提高大鼠对缺氧的耐受力,且呈良好的量效关系。
Objective: To observe Toutongning's effect on function of platelet and rats' tolerance ability of hypoxia, and to explore their mechanisms. Methods: Two hundred Wistar rats, weighing (180±20)g, in both sexes, were divided into four groups at random and were given NS, high and low dosage of Toutongning and ergotamine respectively. The coagulation and bleeding time, hemorrheology index of rats with blood stasis induced by cold , mortality rate of rats induced by ADP and rats' living time under hypoxia were tested. Results: Rats' coagulation and bleeding time in Toutongning group was longer, hemorrheology index of testing group with blood stasis induced by cold was improved, mortality rate of rats induced by ADP was lower and rats' living time under hypoxia was longer compared with control group (P〈0.05). Conclusion:Toutongning can prolong the coagulation and bleeding time, improve hemorrheology index of rats with blood stasis induced by cold, lower mortality rate of rats induced by ADP and improve rats' living time under hypoxia with a dose-dependent manner.
