大肠癌E1AF基因的表达对MMP-9的调控作用

摘要 目的:探讨大肠癌患者血浆基质金属蛋白酶-9(MMP-9)和肿瘤组织中E1A因子(E1AF)表达的临床意义及两者之间的关系。方法:采用酶链免疫吸附法(ELISA)检测51例大肠癌患者血浆MMP-9浓度,采用RT-PCR方法检测肿瘤组织中E1AF的表达,进行定量和半定量分析。结果:Ⅲ、Ⅳ期大肠癌患者血浆MMP-9浓度和肿瘤组织中E1AF的表达水平高于Ⅰ、Ⅱ期患者(P<0.05),Ⅳ期大肠癌患者MMP-9的浓度高于Ⅲ期患者(P<0.05);血浆MMP-9浓度和肿瘤组织中E1AF的表达呈正相关(r=0.77,P<0.05)。结论:患者血浆MMP-9和肿瘤组织中E1AF的高表达与大肠癌浸润转移有关,E1AF参与MMP-9的表达调控。 Objective: To explore the relationship between the expression of E1AF and the concentration of matrix metaloproteinase-9 (MMP-9) in the colorectal cancer. Methods: MMP-9 concentrations in the plasma of 51 cases with colorectal cancers were determined by Sandwich ELISA method. The expressions of E1AF were determined by RT-PCR and quantitatively and half-quantitatively analysed. Results: The plasma concentration of MMP-9 and the expression level of E1AF in stage Ⅲ and Ⅳ of cancer were higher than those in stage Ⅰ and Ⅱ, and the plasma concentration of MMP-9 in stage IV was higher than that in stage Ⅲ (P〈0.05). The expression of E1AF was positively correlated with the plasma concentration of MMP-9. Conclusion: The high concentration of MMP-9 and the expression of E1AF are associated with the metastasis. The level of MMP-9 is regulated by E1AF.
