山楂提取物抑制六价铬对人外周血淋巴细胞致突变作用研究

Antagonistic Effects of Organic Extracts of Hawthorn Fruit on Chromium(Ⅵ)-induced Mutation in Human Peripheral Blood Lymphocytes

摘要 目的:探讨山楂提取物抑制六价铬的致突变作用。方法:采用人外周血淋巴细胞程序外DNA合成(UDS)试验检测了0.03、0.015和0.0075g/m l山楂提取物对重铬酸钾(含C r6+0.31μg/m l)所致淋巴细胞程序外DNA合成的影响。同时检测山楂提取物与重铬酸钾的加入顺序对UDS的影响。结果:阳性对照(重铬酸钾,含C r6+0.31μg/m l)组CPM值显著高于阴性对照(蒸馏水)组,P<0.01。山楂提取物与重铬酸钾同时加入以及两者间隔2h先后加入均显示山楂提取物浓度与CPM值呈显著负相关(P<0.05),在上述不同加入方法中,山楂提取物的各浓度均显示CPM值显著低于阳性对照。结论:山楂提取物具有抑制C r6+的致突变作用。 Objective: To study the antagonistic effect of organic extracts of Hawthorn Fruit on chromium (Ⅵ)-induced mutation. Methods: Using unscheduled DNA synthesis (UDS) assay, the effects of 0.03 g/ml , 0. 015 g/ml and 0. 0075g/ml organic extracts of Hawthorn Fruit on UDS induced by potassium bichromate 0. 31μg/ml chromium (Ⅵ) in human peripheral blood lymphocytes were determined . The effects of the order of adding organic extract of Hawthorn Fruit and potassium bichromate into cultured lymphocyte suspension on UDS were observed at the same time . Results: The value of CPM revealed a significantly higher level in positive control group exposed to 0.31μg /ml chromium (Ⅵ) compared with that in negative control group exposed to distilled water (P〈0. 01). In the comparative experiments with different methods for adding the substances to be tested : simultaneously adding the organic extracts of Hawthorn Fruit and potassium chromate into the cultured lymphocyte suspension , and adding the Fruit first and potassium chromate afterward or in turn at intervals of 2 hours, the values of CPM revealed a significant negative correlation with the exposure levels of organic extracts of Hawthorn Fruit (P〈0. 01),and each value of CPM in all Hawthorn Fruit groups was significantly lower than that of positive control group. Conclusion. Hawthorn Fruit revealed significant antimutagenic effect on chromium ( Ⅵ) .
