摘要
目的:研究γ射线照射对单采血小板保存过程中趋化性细胞因子含量的影响。方法:应用COBE SpectraTM细胞分离机单采血小板,然后以27Gy的γ射线照射,于标准条件下保存5d,间隔取样用ELISA方法检测MCP-3,MCP-1,MIP-1α及RANTES含量。结果:对照组单采血小板在保存1d后,所有趋化性细胞因子含量与保存前比较显著升高(P<0.05),照射组保存3d后,RANTES、MCP-3、MIP-1α含量与保存前比较无明显差异(P>0.05),但在保存5d后,所有趋化性细胞因子均与保存前比较有明显升高(P<0.05)。在保存1d后照射组所有趋化性细胞因子含量均明显低于对照组(P<0.05)。结论:27G yγ射线照射能抑制单采血小板保存过程中趋化性细胞因子含量的升高,从而可以有效地预防单采血小板输注后的非溶血性发热反应。
Objective: To study the effect of prestorage gamman radiation on the chemokine levels in storage apheresis platelets. Methods: Apheresis platelets were prepared by the COBE spectraTM cell separator,then irradiated by gammaray at the does of 27 Gy stored at standard condition for 5 days.chemokine levels were measured by ELISA at various storage time. Results: The contents of all chemokine in apheresis platelets of control group after 1 days storage were significantly different from those of prestorage (P〈 0,05).RANTES,MCP-3 ,MIP-1α in apheresis platelets of irradiated group after 3 days storage compared to those of prestorage showed no significant change (P 〉 0.05), but after 5 days storage, the content of all chemokines were much higher than those of prestorage(P〈 0.05).After 1 day storage the contents of all chem okines in radiation group were lower than that of control group (P〈 0.05). Conclusions: Gamma radiation at 27 Gy is effective in preventing the generation of chemokines in the storage of apheresis platelets.Thus it could prevent monhemolytic febrile transfusion reaction after platelet concentrates transfusion.
