摘要
采用Annexin-V-FITC/PI染色流式细胞术检测细胞凋亡的方法,分别观察Mtb-Ag(10.0μg/ml)刺激特异性激活的人γδT细胞不同时间凋亡情况,并研究Rottlerin(PKC抑制剂)预处理对Mtb-Ag诱导γδT细胞凋亡的影响。结果显示Mtb-Ag刺激3h、6h、12h、24h均可显著诱导γδT细胞凋亡(P<0.01),凋亡细胞比例在44.21%~51.76%之间;Mtb-Ag诱导γδT细胞凋亡可被Rottlerin(40.0μmol/L)抑制,凋亡抑制率为98.6%。提示PKC途径参与Mtb-Ag激发活化的γδT细胞凋亡过程。
The apoptosis of human γδT cells induced by Mycobacterium tuberculosis(Mtb) antigens(10.0μg/ml) in different period of times was observed by flow cytometry(FCM) using the Annexin-V-FITC/PI staining, and the effect of pretreatment with the protein kinase C(PKC) inhibitor Rottlerin on the apoptosis of human γδT cells was also investigated. The experimental results showed that the apoptosis of γδT cells could be induced by stimulation of cells with Mtb antigen for 3, 6, 12 and 24 hours with the apoptotic percentages of 44.21% to 51.76%, and the apoptosis of γδT cells could be inhibited by the pretreatment with Rottlerin (40.0 μmol/L) with inhibition rate of apoptosis of 98.6%. These results suggest that PKC signaling pathway participates in the apoptosis of activated human γδT cells stimulated by Mycobacterium tuberculosis antigens.
