摘要
为观察苏木乙酸乙酯提取物对同种异位心脏移植大鼠移植心肌FasLmRNA表达的影响,并探讨苏木免疫活性部位抗排斥反应作用机制,本研究以Wistar大鼠为供体,以SD大鼠为受体,建立大鼠腹腔异位心脏移植模型,将手术成功鼠随机分为模型组、苏木组、环孢素A组,采用RT-PCR法检测FasLmRNA表达。常规方法观察心肌病理形态学。结果表明,①与模型组相比,环孢素组和苏木组能减轻移植心肌病理损伤(P<0.01);②与模型组相比环孢素组和苏木组能明显下调FasL基因表达(P<0.01),环孢素A组与苏木组相比差异无显著性(P>0.05)。结论:苏木乙酸乙酯提取物能够减轻移植心脏心肌的病理损伤,能够下调大鼠移植心脏心肌FasL的基因表达,这可能是抗急性排斥反应因素之一。
To observe the influence of ethyl acetate extracted from Chinese medicine sappan wood on the expression of myocardial FasL mRNA in acute rejection after allogenic cardiac transplantation of rats, the allogenic cardiac transplantation model was established by using Wistar rats as donors and SD rats as recipients and the rats with successful operations were divided into three groups, i.e. model group, sappan wood group and cyclosporin A (CsA) group. The expression of FasL mRNA was determined by RT-PCR and the pathomorphology of heart muscles was observed by routine methods. It was found that compared with the model group, the sappan wood and CsA groups of rats showed obvious improvement in the pathologic changes in heart muscles, while there was no significant difference between sappon wood and CsA groups. Meanwhile, the expressions of FasL mRNA in sappan wood and CsA group were much lower than that in model group (P〈0.01), but also there was no difference in sappan wood and CsA groups of rats. It is concluded that ethyl acetate extracted from sappan wood demonstrates immunosuppressive effeet through reliving the pathological effects and inhibiting the expression of FasL mRNA in allogenic transplantation of rats.
