摘要
目的:采用 HPLC 法测定芫荽籽中绿原酸的含量。方法:色谱柱为 Hypersil-ODS 柱(6.0 mm×125 mm,5μm),以甲醇-0.4%乙酸(25:75)为流动相,流速为1.0 mL·min^(-1),柱温为室温,检测波长328 nm,进样量5 μL。结果:在上述色谱条件下,绿原酸对照品的峰面积与其质量在0.015~0.106μg范围内线性关系良好,回归方程为 Y=9903.85X+12.72,r=0.9996,平均回收率为98.3%(n=3)。结论:此方法简单快速,结果准确。
Objective:An HPLC method for the quantitative determination of chlorogenic acid in Coriandurm seed was investigated. Method :The sample was extracted with 50% methanol solution and then analyzed by HPLC. The operating conditions were Hypersil- ODS column(6. 0 mm× 125 mm,5μ m)at room temperature,mobile phase of methanol-0. 4% acetic acid(25: 75)at 1.0 mL · min^-1 and UV detection at 328 nm. Results:There was a good linear relationship between the area of chlorogenic acid and the mass of sample in the range of 0. 015 - 0. 106 μg for chlorogenic acid. The corresponding regression equation was Y = 9903. 85X + 12. 72, r = 0. 9996. The average recovery was 98.3% ( n = 3 ). Conclusion:This method is simple, specific and accurate.
