摘要
通过癌胚抗原单克隆抗体(CEAMcAb)免疫家兔获得CEAMcAb抗独特型抗体(Ab_2),Ab_2不但能竞争抑制CEAMcAb与大肠癌细胞HT-29的结合,而且诱导小鼠对大肠癌细胞HT-29的迟发性过敏反应。经过Ab_2免疫小鼠血清能和CEA反应,提示Ab_2能诱导抗CEA抗体(Ab_3)产生,而AB_3又可与CEA表达细胞大肠癌细胞HT~29反应。上述结果提示Ab_2中含有HT-20细胞相类似的抗原决定簇,具有模拟CEA的独特性。
abbits were immunized with CEA McAb against human Colon cancer to produce anti- CEA McAb-idiotyp-ic antibody(Ab_2).The Ab_2 could not only competitively inhibit the binding of CEA McAb to colon cancer cellsHT-29,but also induce delayed-type hypersensitivity(DTH)to HT-29 in mice. The reaction of CEA with serumof mice immunized with Ab_2 suggest that Ab_2 can induce the preduction of Ab_3 which could bind HT-29.The re-sult indicates that Ab2 possesses determinants(internal image antigen)similar to those on HT-29,Therefore,Ab_2could mimic tumor-associated antigen CEA.
