一个抗CEAMcAb独特型抗体建立、纯化和初步鉴定

Development,Purification and Preliminary Functional Study of Anti-idiotypic Antibodies against CEA McAb

通过癌胚抗原单克隆抗体（ＣＥＡＭｃＡｂ）免疫家兔获得ＣＥＡＭｃＡｂ抗独特型抗体（Ａｂ＿２），Ａｂ＿２不但能竞争抑制ＣＥＡＭｃＡｂ与大肠癌细胞ＨＴ－２９的结合，而且诱导小鼠对大肠癌细胞ＨＴ－２９的迟发性过敏反应。经过Ａｂ＿２免疫小鼠血清能和ＣＥＡ反应，提示Ａｂ＿２能诱导抗ＣＥＡ抗体（Ａｂ＿３）产生，而ＡＢ＿３又可与ＣＥＡ表达细胞大肠癌细胞ＨＴ～２９反应。上述结果提示Ａｂ＿２中含有ＨＴ－２０细胞相类似的抗原决定簇，具有模拟ＣＥＡ的独特性。

abbits were immunized with CEA McAb against human Colon cancer to produce anti- CEA McAb-idiotyp-ic antibody(Ab_2).The Ab_2 could not only competitively inhibit the binding of CEA McAb to colon cancer cellsHT-29,but also induce delayed-type hypersensitivity(DTH)to HT-29 in mice. The reaction of CEA with serumof mice immunized with Ab_2 suggest that Ab_2 can induce the preduction of Ab_3 which could bind HT-29.The re-sult indicates that Ab2 possesses determinants(internal image antigen)similar to those on HT-29,Therefore,Ab_2could mimic tumor-associated antigen CEA.
